چکیده

Background Individuals with Phelan-McDermid syndrome (PMS) present a wide range of diagnoses: autism spectrum disorder, intellectual disability, or schizophrenia. Differences in the genetic background could explain these different neurodevelopmental trajectories. However, more parsimonious hypothesis is to consider that they may be same phenotypic entity. Catatonic disturbances occasionally reported from adolescence onwards PMS prompts exploration this clinical entity an early-onset form catatonia. The largest cohort children childhood catatonia was studied by Wernicke-Kleist-Leonhard school (WKL school), which regards as collection qualitative abnormalities psychomotricity predominantly affecting involuntary motricity (reactive and expressive). aim study investigate presence psychomotor signs three young adults carrying mutation intragenic deletion SHANK3 gene through prism WKL conception Methods This designed exploratory case study. Current phenomena were investigated semi-structured interviews parents, direct interaction participants, documents reporting observations participants at other healthcare professionals. Results findings show catatonic manifestations evolved into chronic form, possible phases sub-acute exacerbations starting adolescence. Conclusion symptoms associated autistic traits leads us singular fundamentally related mutations Further studies are needed confirm our observations.

Contribution of SHANK3 mutations to autism spectrum disorder.

Mutations in SHANK3, which encodes a synaptic scaffolding protein, have been described in subjects with an autism spectrum disorder (ASD). To assess the quantitative contribution of SHANK3 to the pathogenesis of autism, we determined the frequency of DNA sequence and copy-number variants in this gene in 400 ASD-affected subjects ascertained in Canada. One de novo mutation and two gene deletions...

Looking at the Schizophrenia Spectrum Through the Prism of Self-disorders: An Empirical Study

Nonpsychotic anomalies of subjective experience were emphasized in both classic literature and phenomenological psychiatry as essential clinical features of schizophrenia. However, only in recent years, their topicality with respect to the construct validity of the concept of the schizophrenia spectrum has been explicitly acknowledged, mainly as a consequence of the increasing focus on early de...

Decalogue of Catatonia in Autism Spectrum Disorders

This article is an unabashed drumroll for increased recognition and treatment of catatonia in autism spectrum disorders (ASD). This new diagnostic and treatment paradigm has emerged during the last decade (1, 2) and is supported by changes in catatonia classification in DSM-5 (3) purporting to boost recognition of pediatric catatonia and catatonia in ASD (4). Findings are summarized, a vignette...

Lithium as a rescue therapy for regression and catatonia features in two SHANK3 patients with autism spectrum disorder: case reports

BACKGROUND Phelan-Mc Dermid syndrome is a contiguous disorder resulting from 22q13.3 deletion implicating the SHANK3 gene. The typical phenotype includes neonatal hypotonia, moderate to severe intellectual disability, absent or delayed speech, minor dysmorphic features and autism or autistic-like behaviour. Recently, point mutations or micro-deletions of the SHANK3 gene have been identified, ac...
